Problems of the Quantitative Determination of Monoclonal Serum Immunoglobulins

Summary:

       The densitometric estimation of monoclonal immunoglobulin concentration in sera of 28 patients was imple- mented using two methods. The first was automated electrophoresis performed with the Hydrasys-Hydragel (Sebia) system, the second was manual electrophoresis on agarose gel. Concentrations of monoclonal immuno- globulins in groups were compared using Student’s paired t-test and no significant differences were found. The concentration of serum albumin was determined by the two mentioned methods and additionally by the bromcresol green photometric method on Hitachi 911. Statistical evaluation of the results revealed differences between the groups. The third goal of this paper was visual evaluation of the very small Ig monoclonal components and of the residual concentrations of monoclonal Ig after therapy. For this purpose the manual proceeding proved better and more sensitive.
